INTENDED USE
For in vitro quantitative determination of Total Bile Acid in human serum or plasma.
CLINICAL SIGNIFICANCE
The assay kit is for determination of serum total bile acids (TBA). Total bile acids are metabolized in the liver and hence serve as a marker for normal liver function. Serum total bile acids are increased in patients with acute hepatitis, chronic hepatitis, liver sclerosis and liver cancer.  
ASSAY PRINCIPLE
Bile acids were specifically oxidized by 3-hydroxysterol dehydrogenase (3-HSD) and oxidized thio-nicotinamide urea purine dinucleotide (thio-nad) to form 3-ketosterol and reduced thio-nicotinamide urea purine dinucleotide (thio-nadh). In the presence of 3-hydroxysterol dehydrogenase and reduced nicotinamide urea purine dinucleotide (thio-NADH), 3-ketosterol was regenerated into bile acid and oxidized nicotinamide urea purine dinucleotide (thio-NAD). As mentioned above, the sensitivity of the test is improved by cyclic release. The concentration of bile acid was determined by measuring the absorbance change of reduced β - nicotinamide urea purine dinucleotide (thio-NADH) at 405 nm.
